Outline of Research at the Start

線維化や肝硬変に対する新たな治療法の開発は、肝疾患を制御する上で最重要課題であるが、未だ有効な治療薬は開発されていない。その理由に、肝細胞障害から肝線維化・肝硬変に至る過程を、培養系で再現できる肝疾患モデルがないことが挙げられる。我々はヒトiPS 細胞から様々な肝構成細胞を分化誘導する技術や線維化の主体である肝星細胞の活性化過程を定量的に評価する技術の開発に成功している。本研究ではiPS 細胞由来の肝構成細胞を用いた肝オルガノイド形成法を開発し、肝疾患モデルへ応用するとともに、肝星細胞活性化の制御機構を明らかにする。

Outline of Final Research Achievements

We established a three-dimensional co-culture system using hiPSC-derived quiescent hepatic stellate cells and human hepatocarcinoma-derived cell line (HuH-7) and demonstrated that hepatic stellate cell activation can be induced after induction of liver injury. We also successfully generated senescent hepatic stellate cells from hiPSCs. We developed a high throughput system to screen compounds that revert the activated hepatic stellate cells to a quiescent-like state.

Academic Significance and Societal Importance of the Research Achievements

静止期肝星細胞をヒト生体から大量に調製することは不可能である。本研究では、ヒトiPS細胞技術を基盤として、一連の培養系で、静止期肝星細胞が活性化し、細胞老化する過程を再現することに成功した。肝星細胞は肝臓における線維化のドライバーであることから、これらの細胞を解析することで肝線維化の病態メカニズムの解明に貢献することが期待される。さらに、肝線維化に対する治療薬の開発にも応用可能であることから、本研究成果の社会的意義は大きい。本研究で同定した活性化肝星細胞を不活化し正常な静止期肝星細胞へと誘導する肝線維症治療薬候補化合物群の開発は、多くの肝疾患克服に貢献することが期待される。
