Outline of Research at the Start

性分化は有性生殖をおこなう生物に必須の生命現象である。ほ乳類では、性決定遺伝子Sryによって性的に未分化な生殖腺がオス化する。私たちは、マウスSryにこれまで未同定だった第2エキソンが存在しており、それが新規性決定因子SRY-Tをコードしていることを明らかにした。（宮脇と立花ら、Science 2020）。本研究では、SRY-Tの機能をさらに追及しつつ、Sry遺伝子の進化の謎も明らかにする。

Outline of Annual Research Achievements

個体が雌雄へと分化すること（性分化）は、有性生殖をおこなう生物に必須の生命現象である。ほ乳類では、性決定遺伝子Sryが胎仔期の生殖腺の体細胞で一過性に発現し、性的に未分化な生殖腺をオス化（精巣化）する。ごく最近私たちは、マウスの性決定遺伝子Sryにはこれまで未同定だった第2エキソンが存在していること、またそれが新規SRYタンパク質SRY-Tをコードしていることを明らかにした。SRY-Tはタンパク質分解を受けないため、生体内で実際に機能している真の性決定因子であった。一方既知の SRY（SRY-S）は、タンパク質分解モチーフ（デグロン）を有している機能不全型のSRYであることが明らかになった。本研究の成果は、これまで30年の長きにわたって信じられてきた、ほ乳類の性決定遺伝子に関する教科書の記載を大幅に書き換えるものであった。これらの新発見に基づいて下記の研究課題を推進した。本研究によって得られた成果は、Sryの作用機序とその進化に関する既成概念の抜本的な見直しにつながる。
1) 真のマウス性決定因子であるSRY-Tのパートナー因子と標的遺伝子の同定:マウス胚性幹細胞を用いた実験により、SRY-Tと相互作用する分子を多数同定した。それらの遺伝子の欠損マウスを作製し、性分化の表現型解析を行った。
2) Sry遺伝子の第2エキソンのゲノム進化過程の考察：野生のオスのハツカネズミ捕獲に成功した。Sry mRNAの配列解析の実験が現在進行中である。
3) 機能不全型の性決定因子であるSRY-Sのタンパク質分解メカニズムの解明：SRY-Sに特異的に相互作用するユビキチンE3リガーゼを複数同定した。SRY-T欠損の遺伝子背景でそれらの遺伝子の欠損マウスを作製し、表現型解析を行った。
