Outline of Research at the Start

カンピロバクターは国内外の主要な細菌性食中毒の原因菌で、鶏肉とその関連食品が原因食品として重要視されている。カンピロバクターを保菌した鶏が食鳥処理場に搬入されると、食鳥と体全体が菌に汚染されるため、その付着メカニズムの解明と制御法の確立が喫緊の課題である。研究代表者らは、菌体側と鶏宿主側に特異的に結合する付着因子が存在するとの作業仮説を立て、両者から複数の蛋白質を検出・同定した。本研究課題では、菌体表面に発現する多糖体などの蛋白以外の付着因子を検索・同定する。そして、菌体が皮膚表面に存在する付着因子と特異的に結合するメカニズムとその生物学的意義を解明する。

Outline of Final Research Achievements

When chickens carrying the foodborne pathogen Campylobacter jejuni are brought into a processing plant, the chickens are contaminated with the bacteria. This research project aims to elucidate non-protein attachment factors on the bacterial side involved in the adhesion of C. jejuni to the chicken skin. We compared the adhesion of C. jejuni strains deficient in capsules (kpsM), phospholipids (pldA), and flagella (flaAB). The number of bacteria adhering to the skin of the capsular-deficient strain (kpsM- and pldA-) was significantly lower than that of the wild-type strain and non-flagellated mutant. Furthermore, when the adherence of the wild strain was examined after the capsules of the wild strain were applied to the skin, the adherence of the wild strain decreased in a concentration-dependent manner. Thus, it was suggested that the capsules of C. jejuni are an adhesion factor for chicken skin.

Academic Significance and Societal Importance of the Research Achievements

カンピロバクターの鶏肉に対する汚染機序として、皮膚と菌体外膜の構成因子間の特異的な付着機序の存在に着目した研究はない。 なぜ鶏肉が本菌によって高率に汚染されているかを新たな視点で解明する点が本研究課題の学術的独自性と創造性を有する研究テーマである。さらに、本菌と食鳥と体皮膚の付着機序が明らかにされ、皮膚への付着を制御する技術開発に繋がれば、食鳥処理場のカンピロバクター汚染の低減が期待できる。特に、殺菌剤を用いないカンピロバクターの新たな制御法に繋がる可能性を有している。より安全な食鳥肉の生産に貢献し、ひいてはヒトにおけるカンピロバクター食中毒のリスク低減への寄与が期待できる。
