Outline of Research at the Start |
オピオイド鎮痛薬は長期使用により副作用および耐性を生じるため、副作用のないオピオイド製剤の開発が喫緊となっている。現在オピオイド耐性、副作用を生じにくいアゴニストの開発として、G蛋白シグナルのみを活性化するバイアスアゴニスト、ならびにオピオイドμ/δ二量体化受容体選択的アゴニストの開発が行われている。 当研究では、北里大学薬学部生命薬化学研究室で合成された新規オピオイド化合物を用いて、アゴニストの特性をさらに上回るμ/δ二量体アゴニスト、およびG蛋白シグナルバイアスアゴニストを同定する。候補化合物を絞り込み、また動物実験を完遂させ、企業への導出および共同研究を行うための基盤を構築する。

Outline of Annual Research Achievements |
がん患者の疼痛管理にはゴールデンスタンダードとしてオピオイド鎮痛薬が用いられるが、これらは長期使用により副作用および耐性を生じるため、副作用の少ないオピオイド新規オピオイド鎮痛薬の開発が喫緊の課題となっている。現在オピオイド耐性、副作用を生じにくいアゴニストの開発として、オピオイドμ/δ二量体化受容体に選択的なアゴニスト、ならびに鎮痛を荷うG蛋白シグナルおよび副作用を惹起するとされるβアレスチンシグナルのうち、G蛋白シグナルのみを活性化するバイアスドアゴニストが有力な候補としてその開発が世界中で行われている。 令和4年度は、北里大学薬学部生命薬化学 藤井研究室において合成された化合物群の中で、μ/δ二量体化選択的かつG蛋白シグナルバイアスドシグナル特性を有すると考えられる新規オピオイド化合物の同定に成功し、同化合物を用いた研究を進めた。次に同化合物が鎮痛効果を有するか否かについて、ポジティブコントロールとしてモルヒネを用い動物実験を行った。その結果、私たちの見出した新規化合物はモルヒネと同等の鎮痛効果を示すことが明らかとなった。今後は依存性ならびに副作用に関する動物実験を、モルヒネをコントロールとして比較解析していく予定である。

Reason
これまでに共同研究先の北里大学藤井研究室で見出されたμ/δ二量体高親和性化合物を中心に、藤井研究室と協力して3Dシミュレーション結果に基づいて設計された新たな化合物を複数作製した。これらのμ/δ二量体高親和性候補化合物の活性確認をCellKeyアッセイシステムを用いて行った。その後、G蛋白シグナルを優先的に活性化するバイアス化合物についても、①μ/δ受容体に選択的かつ②G蛋白バイアスの特性を有する化合物に注視し、Flex Stationアッセイシステムを用いてその選択を行うこととした。今年度は、①、②の特長を併せ持った新規化合物の同定に成功し、また動物実験により、新規化合物はモルヒネと同等の鎮痛効果を有することを明らかにした。次年度は新規化合物の副作用についてモルヒネをコントロールとして動物を用いた依存実験ならびに副作用実験を行っていく。

Strategy for Future Research Activity |
本研究において、すでに報告されたμ/δ二量体化受容体特異的アゴニストと考えられているML335を上回る活性を有し、加えて鎮痛効果を有するG蛋白シグナルを選択的に活性化しかつ副作用を惹起するβアレスチンシグナルはほとんど活性化しない新規化合物の同定に成功した。また動物実験により、本新規化合物はモルヒネと同等の鎮痛効果を有することを明らかにした。今後は、この新規化合物がモルヒネ等が有する依存性や副作用惹起作用などを有していないのかについて動物実験で明らかにし、もし依存性や副作用を引き起こしにくい化合物であることが判明すれば、直ちに鎮痛薬製薬企業と臨床開発を行うための共同研究提案を行う。
