Outline of Research at the Start

HIV感染症の克服のためには、ウイルス特異的CTL反応と体内ウイルス排除の機構のより詳細な解析が必要である。申請者はこれまでにSIV感染ビルマ産アカゲザルエイズモデルを用い、長期複製制御維持に必要なCTL反応の性質を部分的に解明した。しかしながらSIV感染後に複製制御に至るか否かを規定する因子は不明であり、さらなる解析が必要である。本研究ではMHC-Iハプロタイプ90-120-Ia共有SIV複製非制御個体に着目し、エピトープに対する細胞性免疫応答と逃避変異選択の高精細な解析を行い、ウイルス特異的CD8陽性T細胞ドミナンシーとSIV複製制御の関係を考察する。

Outline of Final Research Achievements

Viral mutant selections in seven epitope regions associated with CD8-positive T-cell responses were examined in SIV progressors. Hierarchy of these epitope mutations in sequential selection was indicated. Higher viral load was correlated with enhanced accumulation of viral mutations. This model would be useful for analysis of the determinants for sequential epitope mutation selection, contributing to understanding of virus-host CD8+ T-cell interaction in HIV infection.

Academic Significance and Societal Importance of the Research Achievements

SIV感染複製非制御サルを用い、高精度にウイルスの変異選択出現の感染後週数を同定することで、T細胞エピトープ領域における変異選択の階層性を明らかとした。さらに、感染後早期のウイルス増殖とのちの変異選択の蓄積の関連性を示した。本モデルはエピトープ領域における変異選択順序を決定する要因の解析に有用であり、本研究結果はウイルス複製制御下におけるウイルスと宿主CTLとの相互作用の理解に結びつく基盤情報として重要である。
