Outline of Research at the Start

フィラデルフィア染色体陰性の骨髄増殖性腫瘍(MPN)は、造血幹・前駆細胞の体細胞変異により血球が異常に増加する血液腫瘍である。MPNの根本的治療法である骨髄移植は、治療関連死のリスクや移植不適応症例が多いため、ほとんど適応されない。そのため、発症メカニズムに基づいた、完治を目指す新たな治療戦略の確立が求められている。本研究では、MPN患者の一部に共通して見いだされるCalreticulin遺伝子変異産物によって引き起こされるMPNに対する新規治療戦略の開発に資する、サイトカイン受容体MPLのMPN細胞表面での活性化メカニズムを解明する。

Outline of Final Research Achievements

In this study, we elucidated the significance of N-glycan modification of MPL in MPL activation induced by calreticulin (CALR) gene mutation, found in a subset of Philadelphia chromosome-negative myeloproliferative neoplasm patients. Furthermore, we established an experimental system to evaluate the activity of mutant CALR as an MPL ligand.

Academic Significance and Societal Importance of the Research Achievements

本研究で確立した、変異型CALRのMPLリガンドとしての活性を評価できる実験系は、CALR遺伝子変異を有するMPN患者に対する治療薬のスクリーニングや評価に用いることができるため、MPNの治療薬開発に貢献することが大いに期待される。
