Outline of Research at the Start

代表者はこれまでに膵β細胞におけるインスリンシグナル、特にmTORC1活性が膵β細胞量調節に大きく影響することを明らかにしてきた。しかしながら、mTORC1活性が膵β細胞の可塑性、すなわち分化に及ぼす影響については現在もよくわかっていない。そこで、本研究計画では膵β細胞の可塑性にmTORC1活性が及ぼす影響とそのメカニズムの解明を目的とする。代表者が保有する膵β細胞特異的Tsc2ノックアウトマウス、2型糖尿病モデルマウスおよびヒト2型糖尿病患者の膵組織を用いて、網羅的遺伝子発現解析を通して膵β細胞の分化転換への影響を検討し、その機序解明ならびに改善策の確立を目指す。

Outline of Final Research Achievements

Mice lacking Tsc2 specifically in pancreatic β-cells (βTSC2KO) show decreased pancreatic β-cell volume and hyperglycemia in old age. In islets of βTSC2KO mice, Ptf1a, an acinar cell marker, is expressed from young age, and many amylase-positive cells are observed in old age. Lineage-tracing revealed co-positive cells for amylase and YFP, a marker derived from pancreatic β-cells. These results suggest that increased mTORC1 activity may affect the plasticity of pancreatic β-cells, leading to their differentiation into acinar cells.

Academic Significance and Societal Importance of the Research Achievements

2型糖尿病患者では膵β細胞量が減少しているが、その機序についてはまだよくわかっていない。膵β細胞量減少の機序が解明されれば、糖尿病発症・進展を阻止する治療が可能となり、多くの患者にとっても医療経済にとっても大きな改善点であると考えられる。本研究で代表者は、膵β細胞が腺房細胞に分化転換することを明らかにした。この現象はこれまでに報告されておらず、糖尿病の発症・進展予防において大きく寄与する可能性がある。今後、この現象を防ぐ機序についても検討を行い、実臨床への応用を目指したい。
