Outline of Research at the Start

スタージ・ウェーバー(Sturge-Weber)症候群は、顔面の赤あざと、脳血管奇形に起因する難治性てんかん発作などを特徴とするが、根本的な治療法はなく、発達遅滞が引き起こされる小児難病の1つである。近年、GNAQ遺伝子変異が報告されたが、研究に適した疾患モデルは確立されていない。今回申請者は、ゲノム編集技術を用いてSturge-Weber症候群モデルiPS細胞を作成した。血管内皮細胞に分化誘導した後、オンチップ血管網技術を用いて、その脳血管奇形に対する病態解明と治療薬開発を目指す。

Outline of Final Research Achievements

Sturge-Weber syndrome causes intractable epilepsy and developmental delay due to vascular lesion on the brain surface in addition to port-wine stain on the face. Here, we established a perfusable vessel-on-a-chip model of the vascular lesions by combining human iPS cells and genome editing. Moreover, single cell RNA sequencing allowed us to identify the responsible genes and gene pathways that are specifically altered under perfusion conditions.

Academic Significance and Societal Importance of the Research Achievements

スタージ・ウェーバー症候群は生涯にわたり医療介護支援が必要な指定難病である。近年、原因となる遺伝子が同定されたが、詳しい病態は明らかになっていない。今回の研究では、本症候群のモデルiPS細胞の作製を行い、脈管疾患に対する新しい研究手法としての「微小チップ上の血管網と、その灌流技術」を用いることで、病態の一端を明らかにすることができた。今後さらに本研究手法を利用・応用することで、本疾患のみならず他の脈管疾患の病態解明や治療法開発への発展が期待される。
