| Outline of Research at the Start |
ビタミンCはDNA脱メチル化酵素(TET)の補因子であり、エピゲノムを調節することが最近分かってきた。一方、尋常性乾癬や高齢者に多い皮膚がんなどの皮膚疾患の発症には、DNAメチル化異常のようなエピジェネティクスの破綻が密接に関与すると考えられている。よって、皮膚表皮の分化（角化）におけるエピジェネティクス制御機構の解明は皮膚疾患の予防・治療に必要不可欠である。また、表皮でビタミンCが不足するとエピゲノムにどう影響するか関心がもたれる。そこで、本研究では、表皮角化のエピジェネティクス制御機構及びその加齢変化におけるビタミンCの意義を解明する。

| Outline of Final Research Achievements |
We comprehensively analyzed gene expression changes induced by vitamin C in neonatal rat epidermal keratinocytes under Tet knockdown. We found significant differences in mRNA expression of genes involved in cornified envelope (CE) formation, which is important for skin barrier function. This study suggests that vitamin C may be involved in CE formation through epigenetics.

本研究成果は，皮膚疾患の予防・治療や肌の老化防止に向けたビタミンCの表皮への新たな作用機構を明らかにした．また，将来的に表皮角化エピジェネティクスの理解を基にした皮膚疾患の診断や表皮再生医療の進展などの応用も期待できる．
