Outline of Research at the Start

近年、低温でプラズマを発生する技術が進歩したことにより、生命科学への応用が盛んに行われている。WHOが2020年3月11日に新型コロナウイルス感染症はパンデミックであると宣言して以来、感染症対策は世界中で喫緊の課題となっている。本研究では、感染症拡大抑制に資する低温プラズマ及びプラズマ活性溶液の開発を目指す。具体的には豚コロナウイルスや新型コロナウイルスに対する低温プラズマ照射及びプラズマ活性溶液による不活化試験を行う。更に、ウイルスのエンベロープの構造の変化を調べることにより、エンベロープウイルスに対する低温プラズマ及びプラズマ活性溶液による不活化の分子機構を明らかにする。

Outline of Annual Research Achievements

我々は、これまでの研究でプラズマ活性乳酸リンゲル液（PAL）によるコロナウイルスの殺傷効果や豚コロナウイルスや新型コロナウイルスへの殺傷効果を見出していたので、今年度、PALの更なる成分解析を行った。我々ははばひろプラズマ装置により作製されたPALの有機成分として、酢酸、ギ酸、ピルビン酸、2,3-ジメチル酒石酸、グリオキシル酸を同定していたが、更にグリセリン酸、酢酸エチル、トリカルボン酸を同定し、これらの３つを混合することにより選択的殺傷効果を示すことを明らかにした。更に様々なプラズマ装置により、どのような有機物が生成されるかを調べた。また、名城大学と共同でトリプトファンを含む培養液中の大腸菌に酸素ラジカルを直接照射することにより、大腸菌が殺菌されることを見出し、酸素ラジカルを照射したトリプトファン溶液（間接照射）ではそのような効果が見られないことから、酸素ラジカル照射により生成された短寿命活性物質を原因物質として考えていたが、更にESRによりトリプトファンラジカルが計測されたことから、短寿命のトリプトファンラジカルがその有力候補であると考えた。更にメタボローム解析を行い、解糖系が抑制されていることを見出し、解糖系における中間生成物の合成を触媒する酵素であるGAPDHの活性を調べたところ、トリプトファンを含む培養液中で酸素ラジカル照射した大腸菌ではGAPDHの活性が低下していることが確認された。これらの分子機構を論文としてまとめ、今年度、Environmental Technology and Innovation誌に公表された。
