Outline of Research at the Start

ウィルス増殖を抑制するI型インターフェロンの産出を誘導する核酸分子を構築する。人工的に合成したCG配列を有する非メチル化一本鎖核酸(CpG ODN)はトール様受容体(TLR)9のリガンド分子として機能するが、I型インターフェロンに加えて、炎症性サイトカインも誘導するため、感染症悪化が懸念される。本研究では、pHにより構造を変化させる核酸であるiモチーフ構造に着目し、CpG ODNとTLR9との相互作用を細胞内環境に応じて変化させることにより、I型インターフェロンのみを誘導する分子を開発する。

Outline of Final Research Achievements

We aimed to induce type I interferon in immune cells by inserting an unmethylated single-stranded nucleic acid with a CG sequence (CpG ODN). The results showed that nucleic acid with a CpG sequence in the loop region of the i-motif (i-CpG ODN) alone induced interferon-6 but not interferon-alpha, whereas binding to cationic liposomes by electrostatic interaction induced interferon-alpha.

Academic Significance and Societal Importance of the Research Achievements

本研究では、ウィルス増殖を抑制するI型インターフェロンの産出を誘導する核酸分子を構築した。I型インターフェロンは抗ウィルス活性の役割を担っていることから、ウィルス感染の予防や感染後の感染細胞を除去することができる。本研究の成果は、感染症の予防や治療に応用が見込まれ、人々のQOL(クオリティ オブ ライフ)を高めることに貢献する。
