Outline of Research at the Start

抗原特異的memory Th2細胞形成は、アレルギー性疾患の発症・悪化に主要な役割を果たしている。研究者らはこれまでに、酸化ストレス除去機構の制御因子であるTxnipが、memory Th2細胞形成を介して気道炎症を悪化させることを明らかにした。そこで、本研究では、Txnip欠損マウスを用いて1)酸化ストレス関連遺伝子群の発現変動 2)その上流の転写因子に対するTxnipの働き 3)その転写因子を過剰発現させた際の表現型 を解明することにより、Txnipによる酸化ストレス除去の新規分子メカニズムを明らかにすることで、アレルギー性疾患の新たな治療法開発への分子基盤を構築することを目指す。

Outline of Final Research Achievements

The number of patients with asthma and other allergic diseases continues to increase in both developed and developing countries. The formation of memory Th2 cells from effector Th2 cells that react to antigens plays a major role in the onset and exacerbation of allergic diseases. In this study, we found that Txnip, a regulator of the oxidative stress scavenging mechanism, exacerbated airway inflammation via memory Th2 cell formation. Furthermore, we aimed to elucidate the molecular mechanism by which Txnip promoted memory Th2 cell formation, and to establish a molecular basis for the development of novel therapies for allergic diseases and asthma.

Academic Significance and Societal Importance of the Research Achievements

喘息は根本的な治療法のない疾患であるため、患者数が増加し続けており、年間医療費は日本国内で3,500億円にも達する。喘息などのアレルギー性疾患の病態形成に重要なeffector Th2細胞分化の過程においては、エネルギー産生に伴う酸化ストレスが細胞内に発生するため、抗酸化物質を介した酸化ストレス制御機構が活性化やアポトーシスの抑制に重要である。一方、その後のmemory Th2細胞の形成過程における酸化ストレス制御機構は依然不明なままである。本研究を通じ、memory Th2細胞の形成過程における酸化ストレス量の制御機構を解明することは、新規喘息治療法の開発に繋がると期待される。
