Outline of Research at the Start

Slitrk1は神経細胞の樹状突起伸長に関わる細胞膜貫通型タンパク質で,自閉症や運動性および言語性チックを呈するトゥレット症候群の原因遺伝子の1つである.本研究では骨代謝における骨・骨格筋・骨格筋組織それぞれに発現するSlitrk1の機能を解析し,Slitrk1を基軸とした3者の連関を解明する.

Outline of Final Research Achievements

Slitrk1 is a membrane protein involved in the dendritic extension of nerve cells. Slitrk1 has been reported to be one of the responsible genes for Tourette's syndrome. Tourette's syndrome is characterized by motor tics. Although it has been reported that patients with Tourette's syndrome suffer from a high incidence of bone fractures, no research has been conducted on Slitrk1 in bone. The purpose of this study was to investigate the role of Slitrk1 in bone. It was revealed that Slitrk1 is expressed in bone, and involved in bone differentiation. The mouse deleted of Slitrk1 results in reduced body length. These results suggest that Slitrk1 is important for osteoblast differentiation.

Academic Significance and Societal Importance of the Research Achievements

これまでに骨とSlitrk1の関連に着目した研究は皆無であった.また,神経系と骨に共通して発現する因子Slitrk1を基軸として神経系-骨連関の一端の解明を試みた点で学術的独自性は高い.
本研究が今後さらに発展され,骨におけるSlitrk1の役割が詳細に解析されることで骨代謝における役割が明らかになれば,将来的にトゥレット症候群患者の骨折に対しても予防や治療法確立に貢献できると考える.さらに骨-神経連関の解明にも寄与できる可能性があることから社会的意義も高いと考える.
