Outline of Annual Research Achievements

本研究では「植物膜タンパク質のユビキチン修飾と機能発現制御機構の解明」を目指し,3研究課題に取組んでいる。H28年度は以下のような成果を得た。
植物に特有の膜局在型ユビキチンリガーゼATL遺伝子族は,標的となる膜タンパク質をユビキチン化することで,膜タンパク質の安定化,細胞内局在等の制御に関与する。また,その性質から,細胞膜を介した環境シグナルの受容・伝達制御に深く関わる。申請者は,基幹代謝制御に関与するATL31の発見以来,一貫してATL族の機能解明を進めてきた。本申請では,ATL31による細胞膜上の環境シグナル受容と伝達制御機構の全容解明を目指す。また, ATL31の膜局在性に着目し,細胞の内と外の情報受容・伝達制御にATL31がどのように関与するかについて検討した。
ATL31はユビキチンリガーゼ活性に依存的してC/N応答を制御するが,これまでの解析でATL31の細胞内局在性がユビキチンリガーゼ活性依存的に変化することを発見し,ATL31の細胞内局在性がその機能と密接に関係することが示唆されていた。ATL31のユビキチン化標的を調べ得るためIP-MS解析による相互作用因子の探索を行ったところ,SNAREタンパク質が複数同定された。SNAREタンパク質群はSANRE複合体を形成することで膜融合を促進する膜交通制御の鍵因子である。その中でもSYP61は重要で,受容体やトランスポーター,細胞壁合成装置などの様々なタンパク質の小胞を介した輸送制御の中心を担っている。興味深いことに,ATL31はこのSYP61と相互作用し,ユビキチンリガーゼ活性依存的にSYP61の細胞内局在性を変化させることを発見した。これらの結果から,ATL31がSYP61を介して膜交通に作用することでC/N応答を制御する可能性が示唆された。
