Outline of Annual Research Achievements

これまで我々は、HOIL-1L、HOIP、SHARPINのサブユニットからなるユビキチンリガーゼ複合体(LUBAC)がユビキチンのN末端を介する新規「直鎖状ユビキチン鎖」を生成することで、免疫や炎症応答に中枢的な役割を果たすシグナルNF-κB伝達経路を制御することを明らかにしており、現在その生理的・病理的役割の解明を進めている。
本新学術領域研究でまず我々は、IκBキナーゼの制御サブユニット(NEMO)で見出された直鎖状ユビキチン鎖結合性ドメイン（UBANドメイン）を含有するoptineurin(OPTN)に着目した。OPTNの遺伝子変異は緑内障や筋萎縮性側索硬化症(ALS)を引き起こすが、我々はOPTNの緑内障型変異ではなくALS型変異ではOPTNのNF-κB活性抑制能が喪失することを見いだした。このOPTNのNF-κB活性抑制にはUBANドメインを介して直鎖状ユビキチン鎖に結合することが重要であり、我々は共結晶構造解析によりOPTN-UBANと直鎖状ユビキチン結合の詳細な分子基盤を明らかにした(Nakazawa S. et al., Nat.Commun. 2016)。さらにLUBACは、炎症性サイトカイン(TNF-α)によって引き起こされるアポトーシスの過程で、HOIPがカスパーゼによって限定分解を受け、これに伴ってN末端部の脱ユビキチン化酵素結合部位を含む活性抑制的領域とC末端部ユビキチンリガーゼ領域に分割されること、NEMOに加えてFADDがLUBACの基質であり、アポトーシスの過程でこれら基質タンパク質の直鎖状ユビキチン化が減弱することを見出した(Goto E. & Tokunaga F., BBRC. 2017)。これらの解析からLUBACや直鎖状ユビキチン鎖生成の生理的重要性の一端が明らかになった。
