Outline of Annual Research Achievements

組織浸透性の高い長波長に最適化された新規“無蛍光”赤色蛍光タンパク質を開発し、2光子蛍光寿命イメージングによる組織内神経細胞内分子活性化イメージングに応用することが本研究の目的である。現在までに、赤色蛍光タンパク質を鋳型にし、ランダム変異導入により、吸収はあるが量子収率がほぼ0の色素タンパク質(ShadowR)の開発に成功した。また、変異導入にあたって、側鎖が外側に向いている疎水性アミノ酸をより親水性のものに置換することによって、細胞内タンパク質への非特異的結合を大きく減少させることができた。さらに、2光子蛍光寿命イメージング顕微鏡を用いて、mRuby2とShadowRのペアを各種FRETプローブ(CaMKII, Ras, LOV2)に応用することで発現効率やシグナルの大きさ・安定性が優れていることを確認した。これらの結果をまとめて論文(Murakoshi et al. Scientific Reports 2019)を出版した。
