Outline of Annual Research Achievements

ナイミーヘン症候群の蛋白NBS1はそのC末側の約100aaの狭い領域にDNA修復を協調的に行うための、RAD18,RNF20,ATM,MRE1の4種類の蛋白との結合ドメインが存在し、それにより損傷乗り越えDNA合成、クロマチン・リモデリング、チェックポイント、相同組み換え修復を制御する。本研究では、これら拮抗する制御系がNBS1を介したユビキチン化が「選択的に基質タンパク質を識別」する機構を解析する。
RNF20をノックダウンするとRAD18によるPCNAユビキチン化能も低下した。このことはRNF20によるユビキチン化シグナルとRAF18によるユビキチンシグナル経路が重複している可能性を示唆する。そこで、RNF20のシグナル経路をさらに解析した結果、RNF20はヒストンシャペロンFACTと結合して、DNA二重鎖切断部位に集結することが判明した。これに対してRAD18の集結には少なくとも二種類の蛋白が必要であることがマススペクトルを用いた実験による明らかとなった。また、この二種類の蛋白はDNA二重鎖切断には反応せずに、専ら紫外線損傷だけに反応したことから、RNF20とRAD18のユビキチンシグナル経路は全く独立したものである。両ユビキチンシグナルが交差する原因は、RNF20によるクロマチンリモデリングが紫外線損傷応答にも必要と思われる。一方、RAD18結合ドメイン欠損のNBS1ノックインマウスを作製してその性質を解析した結果、ナイミーヘン症候群の特徴である小頭症は観察されなかった。また、免疫グロブリンのサブセットにはわずかに異常が見られたものの、顕著な違いではなかった。このことから、ナイミーヘン症候群の患者に見られる臨床症状の多くは相同組換えなど他のNBS1機能の喪失に由来すると思われる。
