Outline of Annual Research Achievements

パーキンソン病の病態を明らかにするため、ドーパミン作動性神経細胞に対する神経毒であるMPTP (1-methy-4-phenyl-1,2,3,6-tetrahydropyridine) をニホンザルに投与し、パーキンソン病モデルサルを作製した。また、あらかじめ、大脳皮質一次運動野、補足運動野の上肢領域に刺激電極を埋めておく。覚醒下のサルにおいて、淡蒼球外節および内節に多点記録電極を挿入し、神経活動を記録する。大脳皮質一次運動野、補足運動野を電気刺激することにより、淡蒼球外節/内節の上肢支配領域を同定する。その後、多点記録電極を用い、複数のニューロンから神経活動の同時記録を行って相互相関を調べた。正常サルにおいては、同一の体部位局在から記録したにも拘らず、ほとんどの淡蒼球外節/内節ニューロンが相互相関を示さず、互いに独立に発火していた。一方、パーキンソン病モデルでは、多くの淡蒼球外節/内節ニューロン間で活動の相互相関がみられ、ベータ帯域の共振が生じていることがわかった。L-dopaを静脈内投与すると、パーキンソン病症状が改善されるが、その際、淡蒼球ニューロン活動の記録を行ってみると、淡蒼球外節/内節ニューロン間で観察された活動の相互相関とベータ帯域の共振がほとんど消失していた。これらのことから、淡蒼球ニューロンの同期活動やベータ帯域の共振が、パーキンソン病の症状発現に寄与していることが示唆された。
