Outline of Annual Research Achievements

霊長類、げっ歯類モデルでの脳深部振動と運動制御に関して、以下の研究を行った。
パーキンソン病は、60歳以上の約1%が罹患するなど頻度が多い神経難病である。初期においては薬物治療が有効であるが、数年が過ぎると服用時にジスキネジアとよばれる不随意運動が生じるなど薬剤でのコントロールが難しくなる。ジスキネジアの病態メカニズムを調べるため、病態モデルマウスの大脳基底核から神経活動の記録を行った。まずマウスの内側前脳束にドパミン神経毒である6-OHDAを投与してパーキンソン病モデルを作製した。次にL―ドパを腹腔内に連日投与するとマウスは異常運動を示すようになった。また、予め大脳皮質一次運動野に刺激電極を埋めておいた。パーキンソン病、ジスキネジアの潜在状態、ジスキネジア症状発現時に、大脳基底核の出力部である黒質網様部に記録電極を刺入し単一ニューロン活動を記録し、皮質刺激に対する応答を記録した。正常では大脳皮質刺激によって、黒質網様部ニューロンは早い興奮―抑制―遅い興奮からなる3相性の応答を示す。パーキンソン病の際には、抑制が消失し単相性の興奮が記録された。L-ドパの連日投与により潜在的なジスキネジアの状態では、この興奮がやや抑制されるようになり、実際L-ドパ投与後にジスキネジアを示している際には、大脳皮質由来の抑制が増強しているとともに、遅い抑制が消失していた。大脳皮質由来の抑制は大脳皮質―線条体―黒質網様部路(直接路)を介した信号で運動を惹起し、遅い興奮は大脳皮質―線条体―淡蒼球外節―視床下核―黒質網様部路(間接路)を介した信号で運動をストップする働きがあると考えられるので、ジスキネジアは、運動が容易に起こりやすく、起こった場合にストップが効かない状態と考えることができる。
