【矢冨グループ】
・冠動脈疾患におけるリゾホスファチジン酸（LPA）分子種、マイナーグリセロリゾリン脂質の由来とその病態生理における役割の解明：急性冠症候群では、血漿中において18:2 LPA, 20:4 LPA, 22:6 LPAというLPA分子種が、LPAの産生酵素であるオートタキシン非依存性に増加しており、その産生にマイナーグリセロリゾリン脂質が関与していることをヒト臨床検体において明らかにしてきたが、冠動脈・大腿動脈から得た血漿検体を用いた検討でも確認した。また、冠動脈疾患の病態生理に関与していると考えられるリポ蛋白がこれらリゾリン脂質の産生に関与しているかどうかをin vitroで検討し、リポ蛋白質の酸化・糖化によるリゾリン脂質の変動を確認した。
・癌におけるグリセロリゾリン脂質の由来およびその病態生理への関与の解明：肝癌では、癌の進展に重要と想定されているスフィンゴシン1－リン酸（S1P）は、むしろスフィンゴ脂質からグリセロ脂質への中間体である可能性が示唆された。また、肝癌におけるリゾリン脂質定量、関連受容体・酵素の発現を検討し、リゾリン脂質の肝癌進展における特異的関与を示した。また、胃癌腹水を用いた検討でも、リゾホスファチジルセリンを中心としたグリセロリゾリン脂質の増加、病態への関連を追求する意義を確認した。
【本田グループ】
・アトピー性皮膚炎や乾癬などの皮膚疾患の血清・皮膚組織サンプルの収集を行った。
・マウス乾癬モデルを用いて、炎症進展・制御に関与する脂質の同定とそのメカニズムを検討した。脂質メディエーターの一種であるトロンボキサンの受容体が、乾癬発症に重要であるIL-17の産生を制御し、マウス乾癬モデルにおける炎症進展に関与していることを明らかとした。

Reason
本研究においては、リポクオリティを切り口としたヒト疾患の理解に臨床医（臨床検査医・内科医として矢冨、皮膚科医として本田）の視点・立場から取り組むとしていたが、循環器・動脈硬化性疾患、悪性腫瘍（肝細胞癌など）、皮膚免疫・アレルギー疾患に関して、順調に成果をあげることができ、計画通り進行していると考えられる。
急性冠動脈疾患におけるLPA、癌におけるマイナーグリセロリゾリン脂質、乾癬発症におけるトロンボキサン受容体の関与の重要性を明らかにすることができた。
また、研究倫理委員会の承認の元、各種疾患の血液、組織（癌組織、皮膚など）を中心に、着実に、検体収集を行い、領域全体の臨床検体センターとしての基盤も整えた。

| Strategy for Future Research Activity |
これまで得られた知見を踏まえ、急性冠動脈疾患における血漿不飽和LPAの上昇の機序とその治療医学への応用（佐々木班との共同研究）、肝癌・腹膜癌腫症におけるマイナーグリセロリゾリン脂質の関与に関するin vitro実験を中心に進める。また、肝癌で行った検討を大腸癌でも行う。
乾癬発症におけるトロンボキサン関与に関しては、トロンボキサンがどのようなメカニズムでIL-17産生を制御しているか、培養系をもちいて検討する。またマウスモデルにおける乾癬発症に、脂質の関与が示されたため、ヒト検体を用いて、トロンボキサンを含む脂質の一斉定量を行い、病勢、疾患特異的な因子の抽出を試みる。
