【矢冨グループ】
・急性冠症候群では、血漿中において長鎖不飽和リゾホスファチジン酸（LPA）が、オートタキシン非依存性に増加しており、その産生にマイナーグリセロリゾリン脂質が関与していることを冠動脈・大腿動脈から得た血漿検体を用いた検討にて確認した。また、リポ蛋白質の酸化・糖化、冠動脈疾患の発生母地である糖尿病モデルマウスによるリゾリン脂質、ジアシルリン脂質の変動を確認した。
・肝癌、大腸癌では、癌の進展に重要と想定されているスフィンゴシン1－リン酸（S1P）は、むしろスフィンゴ脂質からグリセロ脂質への中間体である可能性が示唆された。この現象はin vitroにて再現できた。また、肝癌、大腸癌におけるリゾリン脂質定量、関連受容体・酵素の発現を検討し、リゾリン脂質の肝癌、大腸癌進展における関与について疫学的検討およびin vitroの実験にて示した。また、胃癌腹水を用いた検討でも、グリセロリゾリン脂質の増加、病態への関連を追求する意義を確認し、腹膜癌播種モデルマウスでも再現されることを発見した。
【本田グループ】
・尋常性乾癬のマウスモデルを用いて、高脂肪食中の飽和脂肪酸が皮膚血管内皮細胞からの細胞遊走因子発現誘導という新規メカニズムを見出した。一方、オメガ３脂肪酸由来脂質代謝物であるレゾルビンE1は皮膚樹状細胞からのIL-23産生制御を介し、IL-17産生細胞の増殖や活性化を制御することで乾癬発症に抑制的に作用することを明らかとした。またオメガ６脂肪酸由来脂質代謝物であるトロンボキサンが、IL-17産生を促進することで、乾癬病態に促進的作用をすることを見出した。乾癬病変部でトロンボキサン産生酵素は上昇しており、バイオマーカーとしてのトロンボキサンの可能性も示唆された。

Reason
本研究においては、リポクオリティを切り口としたヒト疾患の理解に臨床医（臨床検査医・内科医として矢冨、皮膚科医として本田）の視点・立場から取り組むとしていたが、循環器・動脈硬化性疾患、悪性腫瘍（肝細胞癌など）、皮膚免疫・アレルギー疾患に関して、順調に成果をあげることができ、計画通り進行していると考えられる。
特に、急性冠動脈疾患におけるLPA、癌におけるマイナーグリセロリゾリン脂質、乾癬発症におけるトロンボキサン受容体の関与の重要性を明らかにすることができ、これらの現象の疾患における意義を基礎実験によって解明できつつある。
また、研究倫理委員会の承認の元、各種疾患の血液、組織（癌組織、皮膚など）を中心に、着実に、検体収集を行い、領域全体の臨床検体センターとしての基盤も整えた。

| Strategy for Future Research Activity |
【矢冨グループ】
臨床研究は、平成29年度の研究を継続して行う。また、癌の病態生理へのリポクオリティの関与について検討するため、癌の進展に関係があるとされる物質の発現を検討し、リゾリン脂質や関連蛋白の発現との関係について検討を行う。基礎研究では、平成29年度の研究を継続するとともに、多価不飽和LPAが増加する機序にリポ蛋白の糖化が関係するかどうか、マウスモデルを用いて検討を行う。また、脊髄損傷モデルラットを用い、ATX阻害薬の疼痛疾患への応用について企業と共同研究する。また、臨床検体センターとして、佐々木班（佐々木、青木）、杉本班（村上）との共同研究も推進する。
【本田グループ】
高脂肪食が乾癬病態に及ぼす作用メカニズム、オメガ３脂肪酸由来脂質(レゾルビンE1)の乾癬制御メカニズムを動物モデルにてさらに検討し、作用点を明確にする。またトロンボキサンの乾癬バイオマーカーとしての可能性を、臨床検体を用いて解析する
。また、腸内細菌由来脂質代謝物が乾癬病態に及ぼす作用について、動物モデルを用いて検討する。また食事内容、運動情報データとともに、腸内細菌叢・脂質代謝物解析を進め、疾患制御候補の腸内細菌由来脂質代謝物を少なくとも一つ選定し、標的細胞・作用メカニズムを解析する。
