Research Abstract

分子シャペロンが、基質蛋白質をどのようにハンドリング(操作)しているか、基本的な方式が3つあると思われる。第1は、FtsHやClpBなどで、タンパク質をヒモにしてリングの穴の中を糸通しする場合である。第2は、GroEL/ESなどで、分子シャペロンの分子内空洞の中に変性タンパク質を閉じこめる場合である。第3は、DnaK/Jなどで、変性したタンパク質をつかまえて、はなす、これだけを行う場合である。この特定領域研究で追究するのは(第1)ポリペプチド鎖を一方向に引っ張る糸通しのメカニズム、(第2)変性タンパク質を狭い空洞に招き入れる仕組みとその中での折れたたみ方、(第3)結合と解離の制御、である。
