Outline of Annual Research Achievements

C型レクチン受容体(CLR)は、病原体や自己の構成要素に存在する糖鎖を認識し、免疫応答を引き起こす。Dectin-1は病原体上のbeta-glucanを認識する最も特徴的なCLRであるが、Dectin-1の内因性リガンド(自己認識)は十分に理解されていない。本研究では、ヒトのDectin-1が、血小板上に発現するCLEC-2のリガンドであることを見出した。生化学的解析の結果、Dectin-1はストーク領域が高度にO-グリコシル化されており、ムチン様タンパク質であることがわかった。さらに、ヒトDectin-1のEDxxTモチーフに結合したシアリル化コア1糖鎖がCLEC-2のリガンド部位となることが判明した。ヒトDectin-1をマウスに発現させると、CLEC-2のリガンドとして知られるPodoplaninの欠損に起因するリンパ管形成不全と致死性表現系が解消された。この発見は、自然免疫受容体が生理的なリガンドとして機能し、糖鎖修飾によって生体恒常性を制御する初めての例である。このように、本研究では、C型レクチンファミリーDectin-1が、O型糖鎖修飾を受け、同じC型レクチンファミリーに属するCLEC-2と会合し、器官形成に寄与することを明らかにした。
