Research Abstract

本研究グループは、主に微量な循環細胞(例:癌細胞、造血幹細胞)を主たる標的として、本事象を閉鎖系で捉える為のシステム開発を目指し、研究期間終了までにそのproof of conceptを得る事を目標に据えた。東大医科研グループは、微量な循環細胞を物理学的性状により閉鎖系で効率よく分離するための手法を検討するために、大阪大学金子研究室、名古屋大学新井研究室の協力を得て、数マイクロメートルの幅を持つマイクロ流路における微量細胞の通過時間の測定を繰り返した。末梢血細胞、骨髄血細胞と腫瘍細胞株のマイクロ流路における挙動が異なることを示唆する所見が得られており、次年度は特に腫瘍細胞を含む末梢血検体を用いての検討を進めるとともに、既存の体外末梢血単核球分離装置との併用を念頭に置いた持続処理システムの開発に取り組む。
京都大グループ(分担研究者)では、初年度は、in vitro(体外)において蛍光標識ラベリングした細胞をマウス循環血液中へ移植し、共焦点レーザースキャン顕微鏡と高速イメージングシステム用のCCDカメラを組み合わせることで循環血液中を流れる細胞の蛍光を体外より検出することを目標とした。その際に、購入したフローサイトメーター(3レーザー8カラー仕様Becton Dickinso社、FACSVerse)を用いて、マウスよりサンプリングした血液を同時に解析することで、細胞の検出感度・特異度を比較検討した。次年度は、バリデーションしたシステムを用いて、がん転移の指標として注目される循環腫瘍細胞(Circulating tumor cells)を適切なレベリング下において非侵襲的かつ高感度にて検出するシステムを開発していく。

現在、循環血液中を流れる極少数細胞を検出するための原理開発及びシステムのセットアップに多面的に取り組んでいる。共同研究等を通じて、今後の展開に重要な基礎データが蓄積されつつある。

Strategy for Future Research Activity

今後、東大医科研グループでは転移性腫瘍モデル動物から採取した末梢血単核球を用いて、大阪大・名古屋大グループと共同でマイクロ流路による腫瘍細胞の検出と選別法の開発に取り組む。
京都大学グループではヒト循環血液中の蛍光発現細胞を体外から検出するシステムの開発に向け、マウスを用いた動物評価モデルの構築を目指す。手法としてはマウスの腸間膜毛細血管を露出し、予め投与しておいた蛍光発現細胞がその毛細血管を流れるときに発する蛍光発現を共焦点レーザー顕微鏡及び高速イメージングシステムで高感度かつ特異的に捉える。より深部の血管内を観察できるシステム開発に注視し、研究を推進する。
