Outline of Annual Research Achievements

AID2をベースにしたナノボディ抗体を使って標的タンパク質を認識、分解誘導する、単鎖抗体AID2の開発を進めた。必要なコンポーネントを全てコードするall-in-oneプラスミドを作成し、GFPナノボディを利用して、GFP付加した複数のタンパク質の分解をテストした。その結果、比較的発現量が低いタンパク質であれば、90%以上を分解除去できることが明らかになった。さらにp53やRASに対する単鎖抗体を利用して、内在性のこれら因子を分解できることを見出した。
次にマウスでAID2を利用するために、受精卵を利用してROSA26座にOsTIR1(F74G)の導入を試みたが、うまくノックインされたマウスラインが得られなかった。そこで、ES細胞に導入する方法で進めている。mAID-GFPレポーターをROSA26に導入したマウスラインの作成は成功した。脳内分解に向けた分解誘導化合物のスクリーニングは、20種類近く新規化合物を試したが、5-Ph-IAAを超える分解誘導能をもつものは得られなかった。

Reason

昨年度は単鎖抗体を用いた改良AID2を細胞レベルで検証し、GFP付加タンパク質、p53、RASなどを標的に分解誘導に成功した。AID2のマウスへの応用に関しては、mAID-GFPレポーターを発現するマウスの樹立に成功した。

Strategy for Future Research Activity

単鎖抗体AID2法を細胞レベルでより検証するとともに、線虫、マウスなど動物個体への応用をテストする。マウスに関しては、ROSA26座にOsTIR1(F74G)を導入したラインの確立する。脳内分解誘導用のリガンドに関しては、新規アナログをテストして検証する。
