Research Abstract

本研究では、免疫細胞の動態とその制御機構を解明すると同時に、新たなマルチラベリング臓器間細胞動態評価系を開発し、器官内微小環境におけるリンパ球の状態変化と器官間移動の四次元数量的解析を行うことで、リンパ器官連携の実体を明らかにすることを目的としている。本年度は以下の成果を得た。
1) エフェクターT細胞におけるDOCK8の機能を解析するため、Myelin Oligodendrocyte Glycoproteinを認識するTCRを発現するトランスジェニック(Tg)マウスからエフェクターT細胞を分化させ、マウスに移入する実験系を構築した。この系を用いて、DOCK8エフェクターT細胞はin vitroでは正常に増殖し、サイトカインを産生するにも拘らず、野生型マウスに移入した場合、中枢神経系への浸潤がほとんど認められず、実験性アレルギー脳脊髄炎の発症を誘導できないことを見いだした。
2) マクロファージの遊走過程で、DOCK8がCdc42の活性化を介して、核の動きを制御していることを見いだすと共に、この制御に重要な新たな会合分子を同定した。
3) DOCK8は定常状態でも、形質細胞膜に局在する。変異体を用いた解析から、この細胞内局在にDHR-1ドメインが重要な役割を演じていることを見いだすと共に、この制御に重要な候補分子を同定した。
4) 2回ラベリングが可能なin vivo免疫動態評価系に用いるマウスを作製するために、色変換蛍光タンパク質KikGR、 及びKikGRと異なる波長光源で色変換が可能な蛍光タンパク質を同時に発現するコンストラクトを作製した。作製したベクターをtransfectionして細胞株を作製し、各々の色変換前後の蛍光変化を検出出来ることを確認した後、トランスジェニックマウスを作製した。また、単細胞レベルで遺伝子発現解析可能な評価系を導入確立した。

Reason

樹状細胞やマクロファージ、エフェクターT細胞におけるDOCK8の機能や制御機構の解明に向け、重要な知見を蓄積すると共に、研究分担者と連携して、DOCK8の個体レベルでの機能解析に着手する等、研究は順調に進行している。

Strategy for Future Research Activity

研究は順調に進行しており、免疫細胞の動態制御機構に関して、今後多くの成果が得られると期待される。次年度以降、分担研究者との連携を一層強固なものにし、個体レベルでの機能解析を推進すると共に、重要な成果に関しては、HPやマスメディア等を利用して広く国民に発信したい。
