Outline of Annual Research Achievements

本研究は、1) 免疫細胞の動態とその制御機構を解明すると同時に、2) 新たなマルチラベリング臓器間細胞動態評価系を開発し、器官内微小環境におけるリンパ球の状態変化と器官間移動の四次元数量的解析を行うことで、リンパ器官連携の実体を明らかにすることを目的としている。本年度は以下の成果を得た。

(1) myelin basic protein (MBP) 特異的TCRを発現するTgマウスを用いて、エフェクターTh1・Th17細胞におけるDOCK8の役割を明らかにした。(2) DOCK8がnucleokinesis を制御するメカニズムの解明に取り組み、会合分子を同定すると共に、その機能的重要性を確認した。また、DHR-1ドメインに会合する脂質を同定し、その共結晶の構造解析に着手した。(3) DOCK5欠損マウスがアナフィラキシーショックに抵抗性を示す事を見いだし、Nck2やAktといった分子と会合し、GSK3βのリン酸化を制御することで、マスト細胞における脱顆粒反応をコントロールしていること明らかにした。(4) DOCK2とDOCK5の両者を欠損する好中球において、fMLFおよびPMA刺激によるRac活性化が消失し、遊走や活性酸素の産生、neutrophil extracellular trap (NET)の形成が障害されることを明らかにした。(5) 接触性皮膚炎モデルにおいて、皮膚から所属リンパ節に移行したTregを中心に単細胞レベルでの遺伝子発現を解析し、Tregの免疫抑制因子の発現は階層的に制御されていることを明らかにした。(6) 2回ラベリングが可能なin vivo免疫動態評価系の確立に向け、ROSA locusにCAG-loxP-stop-loxP-蛍光タンパク質のコンストラクトを導入したKnock-in miceを作製した。

Reason

樹状細胞やマクロファージ、T細胞を対象に、DOCK8の機能や制御機構に関する重要な知見を得た。また、DOCK5やDOCK2の新しい機能を同定すると共に、研究分担者と連携して、DOCK8の個体レベルでの機能解析に着手する等、研究は順調に進行している。

Strategy for Future Research Activity

研究は順調に進行しており、免疫細胞の動態制御機構に関して、多くの重要な成果が得られると期待される。次年度以降、分担研究者との連携を一層強固なものにし、個体レベルでの機能解析を推進すると共に、重要な成果に関しては、HPやマスメディア等を利用して広く国民に発信したい。
