母乳中の成長因子の存在とその生理学的意義に関する検討.

Research Abstract

RIAを用いてヒト母乳中のIGFーI濃度を測定した。正常児娩出母体は40例、低出生体重児娩出母体は26例である。IGFーI濃度は、酸エタノ-ル抽出を行ったのちにRIAにて測定した。また、ラット胃粘膜中のprost aglandin(PG)をRIAにて測定し、新生児期の胃粘膜の特徴について検討した。胃粘膜中のPGについては、PGE_2および6ーketo PG_1αを、日令1,7,14,21および成熱ラットについて検討した。以下の結果は、平均±SDにて、IGFーIはng/mg蛋白量にて、PGはpg/mg組織重量にて表示した。
(1)HPLCによる抽出液の各分画におけるIGFーIのRIA活性は、標準IGFーIに一致した。
(2)日令0ー4までの期間において、正常児娩出母体と低出生体重児娩出母体の母乳中IGFーI濃度は、それぞれ2.2±0.3,2.4±0.5と有意の差はみられなかった。この傾向は、日令11まで同様に続いた。
(3)母乳にくらべ牛乳中のIGFーI濃度は、0.6±0.1と低値を示した。また、粉乳中にはその存在を認めなかった。
(4)ラット胃粘膜中のPGE_2は、日令1,7,14,21および成熱ラットにおいてそれぞれ0.7±0.5,14.4±1.3,40.7±10.7,94.2±19.7,257±70と加齢とともに著明に増加した。
(5)ラット胃粘膜中の6ーketo PG_1αは、55±15,38±13,333±118,258±72,583±169とPGE_2と同様な結果を示した。
(6)化学的胃粘膜障害に対するインドメサシンの効果は、成熱ラットで16%に予防効果を認めたが、幼若ラットでは認められなかった。
以上の結果より、母乳中IGFーIは生理学的にも重要な役割を有する可能性が示唆されるとともに、幼若期の消化管自体にも成熱後とは異なった特徴が存在することが明らかになった。
