Research Abstract

As a first step in an ongoing study on correlation of oncogene expression and tumorigenesis in trophoblastic diseases. We determined which oncogenes were expressed in hydatidiform mole and choriocarcinoma cell lines. Southern blot hybridization and Northern blot hybridization were done with eighteenkinds of oncogene probes. No rearrangement and amplification of oncogenes were noted in fresh hydatidiform mole. But, when we checked the localization of myconcogen product at light microscopic level by avidine-biotin technique using the monoclonal antibody in the mole, strong expression of myc product was detected in cytoplasm of molar trophoblast compared with normal villi. This findings indicate us that there is an apparent over expression of myconcogene at the transcriptional level. On the other hand, in choriocarcinoma cell, Southern blot hybridization of eleventh oncogenes revealed no amplification and rearrangement in any of twelfth different choriocarcinoma cell lines. Northern blot analysis revealed moderate to abundant transcripts of five oncogenes.
These elevated transcription of oncogenes at the same time may play an important role in carcinogenesis.
