Research Abstract

ガングリオシド認識のプロ-ブとして結合タンパク質、抗体、分解酵素、転移酵素の研究を行った。
1。病原性大腸菌由来の線毛の付着因子の結合するGM3(NeuGC)は仔豚小腸上皮では生後減少することを見いだした。またfanEとfanFの2つの遺伝子をクロ-ニングした。付着因子遺伝子はfanFでFanE産物はFanF産物と複合体を形成して活性を発現することを明らかにした(内貴)。
2。(1)シアロ糖鎖に対するモノクロ-ナル抗体を樹立した。V_H領域遺伝子をNorthan blottingで解析し、一部の抗体に対しては抗イデオタイプ抗体を作成した(神奈木)。(2)複合糖質糖鎖に対するモノクロ-ナル抗体を用いて人乳から18種のオリゴ糖を分離し構造を決定した。内13種は新しい構造のもので非還元末端にシアリルLe^a構造を有していた(山科)。(3)貝類および昆虫類から単理した2種のマンノ脂質に対する抗体を作成し、それを用いて甲殻類にマンノ脂質が存在することを明らかにした。また南極オキアミから新しい構造のホスホノセレブロシドを発見した(杉田)。
3。(1)糖脂質に作用するエンドグリカナ-ゼのアクチベ-タ-を発見し、精製を行った。これを用いて生理的条件下で糖脂質由来の細胞表層糖鎖を外すことが可能となった(山形)。(2)アミノピリジンで蛍光標識した糖鎖を用いて、2種のパラグロボシド糖鎖に作用するNーアセチルグルコサミン転移酵素を見いだした。直鎖構造を生合成する活性は癌化で上昇していることを明らかにした(谷口)。(3)抗体を用いて糖転移酵素活性を測定する系を開発し、血清中のガラクト-ス転移酵素が乳汁中のものと同一性状であり、癌患者では活性が上昇していることを見いだした(飯田)。
