Research Abstract	細胞増殖と核内リン脂質の関連を探るため、次の2つの側面から実験を行った。第1には複製反応に対するリン脂質の影響について調べた。従来、精製したDNAポリメラ-ゼα,β,γがホスファチジルイノシト-ル(Pl)及びカルジオリピン(CL)により特異的に抑制されることを見出した(Yoshida et al.1988)。又、自律増殖配列ヒトcーmycARSを用いたin vitro複製系に対しPl及びCLが同様に強い抑制を示し、この抑制はこれら酸性リン脂質によるトポイソメラ-ゼIの阻害によるものであろうと考えた。(Umekawa et al.1988)。一方、Plの代謝産物がDNAポリメラ-ゼαを活性化するとの報告がなされたので、Plによる調節機構の存在が強く示唆された。我々はラット再生肝及び正常肝からゲルろ過及びモノQカラム(FPLC,平成2年度本申請により購入)を用いてDNAポリメラ-ゼαの低活性型及び高活性型を分離し、更にそれぞれを精製したところ、粗画分のPlP及びlP_2依存性が消失し、モノQカラムにて酵素と分離した画分を再び加えるとlP_2依存性が回復した。このことはlP_2は特定のタンパクを介してポリメラ-ゼαに作用する事を示す。この補助因子を分離精製し同定する研究が進行中である。第2には、核にPlPを特異的に分解するホスホリパ-ゼC(PLC)及びA_2(PLA_2)の高い活性を確認した。核PLCはFPLCで4種に分れ、そのうちの一型、88KDaのものは腹水肝癌細胞核及び再生肝核に認められるが、靜止肝の核には認められない。本活性は肝再生の過程で細胞周期S期に一致して、一時的上昇が認められ、DNA複製との関連が示唆された(Kuriki et al.1992)。増殖期細胞核に特異的なPLCにつき、大量精製及びCDNAクロ-ニングを行うべく鋭意研究を進めている。
