| Research Abstract |
insulin,IGFー1は卵巣でFSH作用のamplitierとして働き,顆粒膜細胞の機能分化に重要な役割を担う。他方,insulin抵抗性糖尿病患者では高率に多のう胞性卵巣症候群(PCOS)を伴うことが知られる。そこで,正常卵巣でのinsulin受容体とIGFーI受容体の発現態度を卵胞成熟,黄体形成との材連で検討し,PCOS卵巣での知見との比較からPCOS成因への関与を明らかにしようとした。その結果,正常卵巣では顆粒膜細胞にinsulin受容体とIGFーI受容体の局在を認め,両者の局在レベルは卵胞発育と共に増大した。黄体期中期の黄体にはinsulin受容体とIGFーI受容体の明瞭な局在を認めたが,卵胞期黄体ではinsulin受容体とIGFーI受容体の局在は陰性となり,かわって黄体周囲の内質細胞にinsulin受容体とIGFーI受容体の強い局在を認めた。PCOS卵巣では顆粒膜細胞のinsulin受容体とIGFーI受容体の局在レベルは低いことを認めた。このことにより,PCOS卵巣顆粒膜細胞ではinsulin受容体とIGFーI受容体の発現異常がinsulinとIGFーIの作用不全を惹起し,PCOSの成因の一つとなる可能性が示唆された。また,退行黄体の間質化にmsulinとIGFーIの関与が示唆された。
他方,胎盤絨毛トロホブライトの増殖,分化とFGF,IGFーIの関わりでは,トロホブラストは自らEGF,IGFーIを産生,分泌すると同時にEG毛ではトロホブラストの増殖を促進するのに対し,妊娠6週以降ではhCG(α,β),hPLの産生,分泌を増加させトロホブラストの分化機能を促進するという2面的な生物作用を発揮することを認めた。他方,IGFーIは妊娠全期を通じてトロホブラストの増殖促進作用が顕著であることを認めた。
