Research Abstract

ヒトヘルペスウイルス感染症の病態は非常に多岐にわたり、臨床医学上重要な問題となっている。本研究班は、ヘルペスウイルス群の感染発症機構の分子生物学的解明、特にヘルペスウイルス群の構造と、ヘルペスウイルス感染症の発症メカニズムと関連を明らかにすることを目的としている。
今年度の実績として、各研究分担者とも、解析の糸口となる生物学的事象の整理の段階から、一歩進んでHSVの分離株間の病原性および潜伏能力の差異を規定する因子の解明、潜伏EBVのゲノムの再活性化スイッチの機構の解析、末梢血単核球でのHHV6の潜伏感染系の解析等に重点を移しつつ成果を挙げつつある。またこれまで動物実験系がないために解明が遅れていたCMVの病原性についてもマウスのモデル系の導入により新しい展開が期待されてきた。以下に各ウイルスごとの実績の概略を示す。
1.単純ヘルペスウイルス(HSV)の感染発症機構;1)マウスのHSV潜伏感染の各種実験系を確立し、株間の潜伏感染の効率を比較した。また、高温馴化培養細胞においてHSVー2の持続感染系を樹立した(新居)。2)臨床分離gC欠損株のgC遺伝子領域の塩基配列を決定し、コ-ド領域内の点突然変異により停止コドンが生じたことがgCの欠損の原因であることを明らかにした。さらにgCがウイルス感染細胞がADCCによる破壊から免れるのに必要であることを示した(森)。3)胎盤抽出液に抗ウイルス活性を見い出した(清水)。4)ドットハイブリダイゼ-ションによる型別診断のため型特異的断片を同定した(栗村)。5)HSV分離株の末端反復配列の長さの差異と初期遺伝子の構造変化の関連をRFLPの解析により検討した(坂岡)。6)リボヌクレオチド還元酵素欠損ウイルス株の病原性および抗ウイルス剤感受性を検討した。さらにLacZ遺伝子挿入により、プロテインキナ-ゼをコ-ドするUS3領域を不活化した変異株を作成し、親株と神経病原性を比較した(西山)。7)HSV粒子に存在し、パルス電気泳動でHSVゲノムより泳動距離の長いDNAについて解析した(倉田)。
2.EBウイルス(EBV)の感染発症機構;1)EBV関連の種々の病態について解析を進め、鼻腔原発T細胞リンパ腫の組織にEBVゲノムを証明した(大里)。2)潜在EBV活性化に働くスイッチ遺伝子BZLF1の上流にプロモ-タ活性が存在することを見いだした(高田)。
3.サイトメガロウイルス(CMV)の感染発症機構;1)マウスCMVに温度感受性変異株とその復帰株、さらに原株と温度感受性変異株の組換え体を得て、病原性の比較を行った(南嶋)。2)PCR法を用いて、慢性関節リウマチ患者の関節嚢液中にCMVを検出した(古川)。
4.ヒトヘルペスウイルス6(HHVー6)の感染発症機構;1)HHV6ゲノムが末梢血単核球のうち付着性の細胞に急性期を過ぎた後も長く検出され、単球系細胞でHHV6が潜伏感染する可能性が示された(山西)。
