Research Abstract	1.RNA結合蛋白cDNAの単離:葉緑体RNA結合蛋白のRNA結合ドメインの配列をもとに、いくつかのcDNAを単離したが、ほとんど既知の5種のものであった。しかし3種の新しいRNA結合蛋白のcDNAを得た。これらのcDNAから推定される蛋白(RGP-1a,1b,1c)は1個のRNA結合ドメイン(80アミノ酸)をN-端末側に、グリシン・リッチ・ドメイン(61-78アミノ酸)をC端末に持っており、ポリ(G)とポリ(U)に高い親和性を示した。しかしこれらの蛋白はトランジット・ペプチドを持たないため核・細胞質系で作用しているものと予想される。転写産物は、葉、根、花や培養細胞に存在するが、特に根にpre-mRNAが多く蓄積している。いずれの遺伝子に対しても5'スプライス部位の異なるオルタナテイブ・スプライシングmRNAが検出された。 2.作用部位の特定:5種のRNA結合蛋白はいずれも強い配列特異性を示さないことが各種結合実験で明らかになった。 3.RNA結合蛋白遺伝子の解析:RNA結合蛋白cp33遺伝子を2倍体タバコのN.sylvestrisより単離・解析したところ、885bp、1187bpと78bpの3個のイントロンを含んでいた。cp33遺伝子はシングルコピー遺伝子で、その転写開始は開始コドン前方22〜180bpの領域内の少なくとも8ケ所から起こり、TATAボックス様配列が見いだされないことから、この遺伝子はTATA-lessプロモーターを持つと考えられ、ハウスキーピング遺伝子と予想される。
