Research Abstract	安定常磁性種をプローブとするESR法による生体内酸素濃度測定法を確立することを目的としている。 (1)安定ニトロキシドラジカル、活性炭、fusiniteの3種について、プローブとしての適格性を比較検討した。後2者は安定性やシグナルの大きさ、酸素濃度に対する感受性は大きいものの、酸素濃度変化に追随する速度がきわめて遅い(平衡に達するのが遅い)という致命的な欠点がある。これに対し、ニトロキシドは細胞内では酸化還元系酵素により常磁性を失うという欠点はあるものの、酸素濃度変化に従って速やかにその線幅を変化させるので、少なくとも細胞外の生体内酸素濃度の測定には好適なプローブである。特に、完全に水素を重水素で置換したニトロキシドは、シグナル強度も大きく、好適なプローブであると判断した。 (2)マウス個体を対象として、生体内酸素濃度の測定を試みた。TEMPONE-D_<16>の溶液をガス透過性のプラスチックフィルムに封入したものをプローブとして、麻酔したマウスの皮下及び腹腔内に埋め込んだ。LバンドESR装置の共振器内の石英試料管にマウスを挿入し、ESR測定を行ったところ、十分な信号対雑音比で、シグナルを観測することができた。種々の濃度の酸素・窒素混合気を試料管に流して、グナルの線幅変化をピーク高さの変化から観測すると、酸素濃度に応答した変化を観測することができた。
