| Research Abstract |
本年度はHPIV-2の感染性cDNAを作製の為の準備の研究を逐行し、かなりの成果を得た。第1には、HPIV-2のすべての遺伝子、NP、P、M、F、HNおよびL遺伝子cDNAをpVCL19ベクターに組み込むことに成功した。第2としては、膜蛋白遺伝子、[F(膜融合蛋白)とHN(膜融合調節蛋白]をpcDLSRαにそれぞれ組み換えてHeLa細胞で発現させたところ生物活性を確認できた。第3としてはHPIV-2P蛋白遺伝子をYM-1ベクターに組み込み、Recombinant Baculoウイルスを作製し、sF細胞で発現させたところP蛋白が大量に発現され、今後この蛋白を精製し、その生物活性、モノクローナル抗体のマッピングを行う予定である。興味深いことに、P蛋白とともにV蛋白も発現され、edited RNAも見い出されたので、いずれかのステップでEditingが起っているのは確かになった。現在、このBaculoウイルス系で見い出されたEditingの分子機構の解析に全力を挙げている。第4としてはHPIV-2M遺伝子をブルースクリプトIIベクターに組み換え、大腸菌で発現したところ、培養細胞でのM蛋白と同分子量のprutuetの検出でき、モノクローナル抗体の反応性も確かめられた。今後、この系を用いてM蛋白の機能を解析して行く予定である。第5としてはRNAVプリコンを作製し、リーダー配列、トレイラー配列、R1配列およびR2配列の機能を解析しており、R1配列がmRNAへの転写に必須であることを示唆する結果を得ている。 さらに、ゲノムサイズのcDNAを構築し、ワクチニアベタターから細胞内発現させた。T7RNAポリメラーゼで全構成蛋白を発現させてウイルス粒子を形成させるために、ブルースクリプトIIベクター系での発現用クローンを作製中である。
