YACベクターを用いたヒトHLA抗原遺伝子領域の発生分化に関与する遺伝子の同定

Research Abstract	ヒト主要組織複合体であるHLA遺伝子群は、自己-非自己の識別に関与し、免疫応答の誘導に深く関わる白血球抗原をコードする多重遺伝子族に属している。我々のグループはすでに、4,000kb中2,200kbをコスミドベクター及び、最近ではYACベクターを用いてクローニングを行っていたが、本年度はHLA-DP遺伝子よりセントロメア側でマウスイノヒ領域に相当し、発生分化に関与すると推定される領域、300kb型並びにクラスII遺伝子とクラスIII遺伝子間領域300kbについて、新たにクローニングを行い、8個の新遺伝子を同定した。このうち、最もセントロメア側に位置する遺伝子は、分裂時の染色体の移動に関与していると考えられる、キネシン様の蛋白をコードしていることが明らかになった。また、クラスII遺伝子とクラスIII遺伝子間領域には、グリコシル化蛋白レセプターをコードする遺伝子も同定され、感受性遺伝子がこの領域にマップされる若年性糖尿病との関連が注目された。この領域には、その他にホメオボックス蛋白のひとつであるPBX3様遺伝子と分化や細胞周期の調節に関与しているint-3様遺伝子が同定され、発生や分化への関与が注目された。このラクスII遺伝子とクラスIII遺伝子間領域はGCrich領域からATrich領域への分節点が存在することでも興味ある領域であり、この領域がクローニングされたことにより、この分節点の詳細な構造やGC対AT比の変化の生物的意義も明かにしうるであろう。発生と分化に関与すると考えられる上記2つの遺伝子に関する機能解析やこの近辺に発症遺伝子がマップされる疾患(ナルコレプシー、強皮症、重症筋無力症など)との関係の検索は現在おこなっている。
