Research Abstract	超硬合金製造の職域集団において、血清中の総IgE値およびCobalt-specific IgE値をアレルギー反応の指標として、ライフスタイルおよびCobalt暴露の複合影響の解析を行った。Cobalt-specific IgE値はCobalt暴露の影響が大きく、一部喫煙の影響もみられた。一方総IgE値はCobalt暴露およびライフスタイルの両方の複合影響を受けていた。また総IgE値が400以上の高値を示す者はとくにこれら諸環境要因の影響を強く受けていた。このグループに属する者では、アトピー等の遺伝要因の関与の可能性も考えられる。アレルギーの遺伝要因としては、HLAのDQ遺伝子の関与が報告されているが、最近遺伝性の強いアトピー患者において11番染色体FCERIB遺伝子或いはその近傍の遺伝子の関与を示唆するデータが報告されている。我々は、この遺伝子の多型情報の豊かなマーカーを用いて、日本人家系における連鎖の解析を開始した。またアトピー遺伝子の社会医学的スクリーニングの準備段階として、極微量血液を用いた場合の最適な保存条件、DNA抽出法、さらにPCRの方法等について、様々な検討を加えつつある。
