Research Abstract

The purpose of this research project is to establish the cultivation methods for Genus Angiostrongylus. Recently, we successfully cultured A.costaricensis(Ac) from the third stage larvae to young adults in chemically defined medium. Nutritional requirements of Ac were examined by deletion of single components from this defined medium. Ten amino acids, choline cloride and glucose were shown to be essential for their development. This is the first study to identify the essential nutrients for development of mammalian parasitic nematodes. The essential component in red blood cells (RBCs), which was essential nutrients for the development of young adult worms of Ac, was examined, and although the investigations were now continued, at least, hemoglobin free ghost and hemin did not support their development. On the other hand, in the cultivations of A.cantonensis, the third stage larvae were cultured to the late third stage, and the 4th stage larvae recovered from rat brain successfully cultured to young adults in same defined medium successfully cultured Ac. Using this culture system, drug screening was performed against the third stage larvae of A.cantonensis, ammnonium salt of embelin, one of the crude drug, were found to have strong antiparasitic effects. In the course of the development of Ac in chemically defined medium, the metabolites were collected, purified the proteins from the metabolites and examined for their antigenic specificity against Ac. However, we could not get the result within the research term. These investigations are now continued.
