Research Abstract	人類は、科学の進歩とともに、膨大な数の化学物質を地球上に造り出したことにより、人類の歴史上、これまで出くわしたことのない物質に曝露されることになった。本研究は、地球レベルでの情報収集と、最新の科学技術を用いて新人為起源有害物質の継世代的影響を総合的に研究することにより、人類の未来をうかがう研究成果を蓄積し、継世代人体障害の防護の資料とするのを目的とする。本年度は、各分担者が、Dioxin専用の飼育装置の設置、検出方法の決定、いくつかの新人為起源有害物質での試行を行い、以下に示した成果を得た。 1.ヒト精子授精能障害の検出(石井): 合成洗剤は0.02%以上で授精能を100%阻害、ダイオキシンは1μg/mlで30%阻害。 2.アルキル化剤(MMS,TEM)によるヒト精子染色体異常の検出(上口): 切断型異常は直線的、交換型異常は二次曲線的に増加。 3.ダイオキシンのマウス精子、卵子への蓄積とDNA損傷の検出(井上): ^3H-TCDDは少量ながら精巣から検出、電顕的にセルトリ細胞の小胞体の退化が認められたが不定期DNA合成は微少。 4.ミニサテライトを用いた継世代的DNA変異の検出(木南): Pc-1プローブを用い、サザン法によるDNA変異検出法を確立。 5.除草剤による生化学的機能障害の発現(藤井): 除草剤のうちグルホシネートはラットに対し、強い海馬刺激と大脳辺緑系刺激および内分泌系障害を誘発した。 6.雄マウスへの作用によるF_1の致死と奇形(野村): エチルニトロソウレアを父マウスに投与すると、F_1に流産、奇形、仮死が有意に増加。 7.ヒトF_1疫学調査(野村): ベトナムでの父親ダイオキシン被曝による子供の奇形発生についての情報収集を行った。
