Research Abstract	本年度は、各種平滑筋の収縮機序の生理学的性質と、収縮蛋白系の生化学的性格を比較し、機能との関連を明らかにすることを目的として実験を行った。Tonic筋の例として、大動脈ならびに気管平滑筋、phasic筋の例として胃、大腸、および子宮平滑筋を用いて実験を行った。その結果、様々な成績が得られたが(裏面の業績目録参照)、本報告書では、特に興味ある知見が得られたニワトリ筋胃平滑筋の成績を紹介する。ニワトリ筋胃は、高濃度Kやアセチルコリンなどいずれの刺激によっても一過性収縮のみを発生する典型的なphasic筋であり、持続性の反応を示す他の筋とは性質が大きく異なる。Fura-2を用いて細胞内Ca濃度と収縮張力を同時に測定すると、細胞内Ca濃度も張力と対応して一過性であった。刺激が継続するにも関わらず、細胞内Ca濃度の増加が一過性に終わる理由として、筋胃のCaチャネルが、電位依存性および細胞内Ca濃度依存性に不活化されるという特異な性格を持つチャネルであることが明かとなった。さらに、収縮蛋白系に関しては、他の多くの平滑筋でみられる受容体刺激による収縮蛋白系のCa感受性増大機構が観察されず、またサイクリックAMPやサイクリックGMP依存性キナーゼによるCa感受性低下もみられなかった。以上のように、phasic筋の典型例であると思われるニワトリ筋胃は、Caチャネルの性質、さらに収縮蛋白質のCa感受性の制御機構について他の筋とは大きく異なることが明かとなった。
