Research Abstract	細菌性スーパー抗原はトキシックショック症候群、猩紅熱、さらにエルシニア感染症の病原性外毒素である。これらの外毒素はジフテリア外毒素のような標的細胞に対する直接的障害作用(細胞溶解や機能障害)はもたずにMHCクラスII分子に結合してTCRVβ選択的に莫大な数のT細胞(それぞれのスーパー抗原について全T細胞の3〜30%)を一括して活性化する。上記感染症における異常反応はこれら外毒素のT細胞活性化作用に帰因するサイトカインの大量産生によって惹起されると考えられる。我々は、平成5年と6年の2年間の研究において下記の研究成果を得た。 1.エルシニア感染症の起炎菌であるY.pseudotuberculosisから新しいスーパー抗原性外毒素Y.pseudotuberculosis-derived mitogen(YPM)を発見した。その遺伝子解析からDNA配列とアミノ酸配列を決定した。YPMは131個のアミノ酸残基からなる分子量14,529ダルトンである。 2.YPMはガラクトサミンとの併用投与によってマウスに対してCD4^+T細胞依存性に致死活性を示す。この致死性にはT細胞産物のTNF-αが関与しているらしい。 3.ブドウ球菌由来スーパー抗原staphylococcal enterotoxin A(SEA)はガラクトサミンとの併用投与によってはマウスに対してCD4^+T細胞依存性に致死性を示す。致死性に関与するサイトカインはTNF-αである。IFN-γの関与については現在解析中である。 4.新生児の免疫学的状況を解析するために、臍帯血T細胞のスーパー抗原,toxic shock syndrometoxin-1(TSST-1)に対する応答性を調べたところ、臍帯血T細胞は成人末梢血T細胞と異なり、免疫記憶の成立が弱いことが明らかになった。
