網膜色素変性症における光受容細胞特異的レチノール脱水素酵素遺伝子の解析

| Research Abstract |
我々は、網膜色素変性症の原因を解明するために、視サイクルの中で特に視細胞に特異的なレチノール脱水素酵素の働きに注目し、ビタミンA代謝の鍵酵素である視細胞特異的レチノール脱水素酵素を精製し、抗体を作成し、この抗体を利用してcDNAを単離し、その構造解析を行ってきた。その結果、(1)精製したレチノール脱水素酵素のN末端から一部の一次構造を決定、(2)ウシ網膜cDNAライブラリーからラット抗ウシレチノール脱水素酵素を用いてレチノール脱水素酵素のcDNAを単離、(3)単離したレチノール脱水素酵素cDNAの塩基配列の決定を終了し、1017個の塩基配列を持つことが判明した。
我々は分離したこのcDNAのレチノール脱水素酵素活性を検討するため、動物細胞用ベクターpSVLにこのcDNAを組み込み、COS7細胞に導入したが、活性の測定ができなかった。大量に材料を得るために、CHO細胞に導入して安定にレチノール脱水素酵素を発現する細胞を得たが、まだクローニングの最中で活性の測定が終わっていない。
