内分泌性神経終末における微小胞の本態とその生理的意義

Research Abstract	視床下部下垂体系の神経分泌ニューロンの神経終末において、ホルモン含有暗調顆粒と共在する小型の明るい微小胞の本態を明らかにするために、種々活性物質の免疫電顕的観察を行った。その結果、ラット視床下部下垂体系の神経終末の微小胞に次のような活性物質の免疫活性が観察された。 1.グルタミン酸様免疫活性抗グルタミン酸抗体を用いてLowikryl K4M包埋後金コロイド法によって観察した結果、下垂体後葉の神経分泌終末において明るい微小胞の密集した領域にグルタミン酸様免疫陽性反応が見られた。 2.グルタミン酸レセプター様免疫活性 NMDA-RI型グルタミン酸レセプター免疫陽性ニューロンは視床下部室周囲核に観察された。またその神経終末では明るい微小胞の周囲にNMDA-RI様免疫活性が観察された。 3.チロシン水酸化酵素(TH)様免疫活性視床下部(A12,A14)のカテコールアミン含有ニューロンの神経終末内に存在する明るい微小胞の密集部にTH様免疫活性が観察された。 4.その他、下垂体後葉系神経終末内の明るい微小胞にシナプス小胞の構成タンパクであるsynaptophysinの免疫活性を観察している。以上、神経分泌終末内に存在する明るい微小胞にはグルタミン酸やそのレセプターが局在している可能性が示唆され、ホルモン分泌に際してautocrine的働きをしているものと推定された。
