Research Abstract |
転写制御の本質は、各種転写因子の作用によって、RNAポリメラーゼが転写対象の遺伝子の選択巾を変えることによるものであることが、示唆されている。その理論の検証と、その分子機構の解明を目指して、先ず、転写因子の作用点の同定を試みた。その結果、大腸菌RNAポリメラーゼでは、αサブユニットのC端領域とσサブユニットのC端領域に、それぞれ一群の転写因子との接点を見出した。しかも、各因子との接点は、それぞれ約10アミノ酸残基のエピトープサイズであった。この発見によって、転写制御の本質に一歩近づくことができた。そこで、この転写因子接点を含む構造ドメインの構造解析を開始した。なお、同様の研究を、真核生物のRNAポリメラーゼIIとウイルスのRNAポリメラーゼにも広げて開始した。
