Research Abstract

RecA蛋白質は,遺伝子組換えの他,遺伝子の発現調節,DNAの複製にも大きく関わるが,その機能発現機構には未解決の現象がまだ数多く残されている。そこで本研究では,進化的に大きく異なる古細菌・原核生物・真核生物のRecA蛋白質の比較解析から,その機能発現機構を解明することを目的として,まず耐熱性菌・古細菌のrecA遺伝子のクローニングを試みた。
まず,ロシア国内の温泉から多くの熱耐性菌を採集し,RecA/RAD51で高度に保存されているアミノ酸配列を元に作製したプライマーを用いて,ゲノムDNAの遺伝子バンクから,recA/RAD51遺伝子をクローニングした。その結果,全ての生物種に共通で,かつ機能発現に必要な領域を明らかにした。また,トポイソメラーゼと有意な相同性を示す新しい遺伝子も見出された。この遺伝子は,古細菌における組換え機構を考える上で非常に興味深いものであり,現在解析中である。次に,全ての生物種に共通する組換え機構の基本的性質を明らかにするために,遺伝子操作系が確立した生物の中で,最も高温で棲息する代表的な耐熱性原核生物の高度好熱菌Thermus thermophilus HB8からrecA遺伝子をクローニングした。遺伝子操作を利用して量産化後精製した好熱菌RecA蛋白質は,DNA鎖交換反応,DNA依存性ATP加水分解などの活性を有しており,これらが相同的組換え反応に必須の性質であることが示された。安定な好熱菌RecA蛋白質を用いてX線結晶構造解析のための結晶化を行ったところ,容易に結晶を得ることができた。DNA存在下での結晶化にも成功した。これらについては現在回折データを収集中である。さらに,好熱菌酵素は一部のドメインだけでも構造を保持できることから,NMR測定を可能にするため,分子同士の会合が起こらない欠失変異体を作製した。今後,これらの構造解析研究によって,RecA蛋白質の機能発現に必要な立体構造および反応に伴う構造変化が明らかになるものと期待できる。
