| Research Abstract |
SM/JとA/J系を祖先系統とする26系統からなるSMXAリコンビナント近交系(RI)を確立しマイクロサテライトDNAをマーカーにRI系統間分布表(SDP)を拡充し、RI系統間に存在する腫瘍、奇形発生率、薬物耐性、感染性、行動、体重などにおける特性の違いを明らかにしていくための基礎調査を行った。一方で、受精卵凍結による保存法の検討を行った。
1.SM/J,A/J及びRIの26系統の兄妹交配による系統維持及び実験供試用の拡大生産を行いながら以下の形質について基礎調査を実施した。
(1)体重及び臓器重量:70日齢にて体重測定後、麻酔死の後解剖し、脳、肺、心、肝、腎、副腎、脾、生殖器(雄:精嚢腺、精巣、精巣上体、 雌:子宮、卵巣)の重量を測定した。RI各系統雄10匹、雌10匹を目標に件体数を増やしているところである。
(2)寒冷適応形質:RI各系統について造巣行動、成長速度、体サイズ、褐色脂肪組織、腎周囲脂肪組織の測定を行っているところである。
(3)血液性状:各RI系統について70日齢において、赤血球数、白血球数、ヘマトクリット値、ヘモグロビン濃度の測定を行っているところである。
2.SM/JとA/Jの間でのマイクロサテライトの多型を検索し、多型が見つかったマーカーについてRI系統間でタイピングを実施した結果、全染色体にわたってマーカーが均等に分布するSDPが得られた。
3.各RI系統の受精卵凍結保存の方法を検討した。幼若マウスへの精腺刺激ホルモン処理による過排卵誘起方法では良い成績が得られなかったので、adult同士の自然交配による受精卵の凍結保存を行うことにした。
4.量的形質の遺伝的解析用コンピューターソフト、マップメーカー/QTLを用いて血液性状のデータの入力を行い解析中である。
