Research Abstract	グローバル化・ボ-ダレス化する世界においてますます問題になってくるのが感染症であり、その予防を含めた対策が必要となってくる。殆どすべての病原性微生物が消化管、呼吸器、生殖器に代表される粘膜に被われた組織から侵入してくることを考慮すると、これらの部位に第一線の防御バリアーを構築することが必要である。そこで重要になってくるのが免疫学の領域で新しいパラダイムを築き上げつつある粘膜免疫機構である。本研究計画では粘膜免疫のユニーク性と重要性に早い段階で注目していた日米の当研究代表者を中心とした若手研究者のグループにより粘膜ワクチンの実現化へ向けた基礎的研究を活発に推進してきた。本年度はTh1型免疫応答調節サイトカインとして注目されているIL-12の粘膜免疫調節因子としての作用について検討し興味ある結果が得られている。ワクチン抗原(破傷風類毒素:TT)とコレラ毒素(CT)を混合ワクチンとして経口免疫すると消化管関連リンパ組織(例、GALT)を中心として抗原特異的Th2型細胞IgAB細胞が誘導されてくる。さらに全身系にもIgGを中心とした抗原特異的免疫応答が誘導されてくる。つまり、経口免疫により粘膜系と全身系両者に抗原特異的免疫応答が液性免疫を中心として誘導する事が出来る。しかしながら、ウイルスに代表される細胞内寄生感染を考えたときTh1型細胞やCTLによる細胞性免疫の粘膜免疫担当組織への誘導も必要である。そこでIL-12をド-タップ型リポゾームにより経粘膜投与しその効果を検討した。粘膜免疫調節因子としてIL-12を併用した場合、期待したように抗原特異的Th1型応答が粘膜系と全身系に誘導された。さらに興味ある点は、IgA誘導に択的に関与するIL-5,IL-6,IL-10を産生するレベル2といわれるTh2型細胞の惹起が維持されていた。つまり、IL-12を粘膜免疫調節因子もしくは粘膜アジュバントとして使用した場合、Th1型応答とレベル2Th2型主導の分泌型IgA両方の免疫応答が誘導出来る可能性が示唆された。粘膜バリアーに細胞性と液性免疫両方を誘導する粘膜ワクチンの開発に向けて貴重な知見である。これらの興味ある結果を含めて粘膜免疫システムの解析を主体に平成9年度には12編の原著論文と9編の総説を本国際協同研究計画を介して国際学術誌に掲載する事ができた。
