Research Abstract	本年度は以下の5つの方向から研究を進めた。(a)生育に必須なrfc3^+遺伝子を見つ、その温度感受性変異株を樹立した。そのうちrfc3-1はUV,MMS,HUに感受性で、制限温度でFACS異常を示し、DAPI染色では核分配が異常な細胞が多かった。これらの結果はRfc3がS/Mチェックポイントに関わっていることを示唆した。この変異株のRfc3では機能発現に重要なDomain VIIIでArgからTrpに点変異していた。(b)出芽酵母にはGIN4と呼ばれる構造上NIK1と類似な遺伝子がある。gin4遺伝子破壊株は致死ではなく、FACS解析では細胞周期に顕著な影響はみられないものの細胞形態に異常を示す細胞が観察された。GFP-Gin4はbud-neckに局在し、Gin4を大量発現させると弱い生育阻害を起こし、細胞の形態がおかしくなった。cdc28変異株とGIN4との二重破壊株が異常に伸長した形態を示したことからGIN4もNIK1同様にDNA複製と細胞質分裂の連動を調節していると結論した。(c)ヒト細胞においてDNA複製開始の制御遺伝子である6種類のHsMCM遺伝子を単離し、抗体も準備してHsMcm2-7複合体全体の機能、構造などを解析した。(d)サイクリンG1とサイクリンG2ノックアウトマウスを作製する準備段階として両方の遺伝子をクローニングして構造を決定するとともに、染色体座位を決定した。(e)マウス・メラノーマ亜株B16-F10(低転移型)とB16-BL6(高転移型)との融合細胞の作製を行って、BL6が示す高転移性が優性な表現型であることを示した。次に両者の差分化cDNAライブラリーを作製して癌細胞に浸潤・転移能を付与する候補遺伝子を多数単離して機能解析してきた。
