Research Abstract	デルマトポンチンは最近見出された新しい細胞外マトリックス成分でプロテオグリカンや細胞接着能を有し、細胞-細胞外マトリックス相互作用やマトリックス構築に重要と考えられている。本研究では肥厚性瘢痕、強皮症の皮膚培養線維芽細胞のデルマトポンチンの遺伝子及びタンパク質発現を検討した。肥厚性瘢痕由来の線維芽細胞のmRNA発現とタンパク質合成は同一患者の正常皮膚由来の線維芽細胞に比べ低下し、強皮症線維芽細胞でも有意に減少していた。サイトカインやマトリックスコラーゲン下での発現を検討したところ、TGF-β1は発現を亢進させ、IL-4は低下させた。コラーゲンをコートした条件下でも、その発現は減少し、コラーゲンの三次元培養条件下で著しく低下した。これらの結果はデルマトポンチンは瘢痕、強皮症の線維化に関連し、デルマトポンチンのコラーゲンの三次元構造やサイトカインの効果は組織の再構成や線維化に重要であることを物語る。
